Timeline Mitopure Softgels

Verdict
"One Bio Star, by founder's ruling. Mitochondrial science translated with uncommon discipline, and stopped exactly where its evidence stops. Mitopure distinguished itself through a proprietary purified molecule at a dose that was itself randomised, predictable human exposure six times that of pomegranate juice, a fifteen-year research programme that has carried one biological proposition from a discovery paper through phase one, two randomised trials, an immune trial, a systematic review and a meta-analysis, a reassuring safety record, NSF certification of the exact softgel sold, and a two-softgel protocol that asks nothing of the user. The anchors computed Finalist: institutional score 3.52, floored at 2.71 by Stewardship, where the company publishes nothing about the container, the waste stream or its footprint; and Efficacy at 3.71, because both major trials missed their primary endpoints and the pooled functional effect is directionally favourable and statistically inconclusive. The ruling covers documentation, not science. No score was moved. The mechanism has left the diagram and entered randomised human investigation. It has not reached its destination."
Most longevity supplements begin with a molecule. Mitopure begins with a biological process. Mitophagy is one of the cell's quality-control systems: damaged mitochondria are identified and removed so the network can renew itself, and its deterioration with age is consequential. Urolithin A became interesting because it appeared able to act on that machinery, and Timeline's proposition is unusually direct: rather than asking the consumer's microbiome to make the molecule from pomegranate, and only about forty percent of people can, supply the molecule itself at a standardised dose. The mechanism is elegant. The question before the Inspectorate was whether the human evidence had become equally convincing. The answer is that it has become measurable and not yet consistent. Exposure is predictable. Plasma acylcarnitines fall and mitochondrial gene sets in muscle shift after four weeks. Hamstring strength rose twelve percent against placebo at the dose sold. And the six-minute walk and ATP production in older adults did not improve, peak power in middle-aged adults did not improve, and five trials pooled together give a walk difference whose confidence interval crosses zero. The signal is increasingly difficult to dismiss. It is not yet consistent enough to call definitive. The most impressive thing about Mitopure may not be any single result. It is the willingness to keep testing the same biological proposition as the questions become harder. What holds the file below a Star on the anchors is not the science, which is a signal, but the paperwork: three purity figures, four patent counts, three trial counts, a GRAS letter described as a declaration of safety, and nothing at all about the container or the footprint. The founder's ruling lifts the computed Finalist to One Bio Star for the molecule, the dose and the programme; the distance to a Star earned outright is a page about the object.

13-Circle Assessment
Inspector Notes
“Mitopure begins with an unusually elegant idea: if the microbiome cannot reliably make the molecule, give the body the molecule directly.
“Urolithin A is not another ingredient pulled from the longevity lexicon and placed into a capsule after the fact. Its development has been accompanied by a fifteen-year research programme examining the biology that made the molecule interesting in the first place, at a Swiss university, by the scientists who then founded the company. That distinction matters, and it is the reason the Inspectorate read every paper rather than the brochure.
“The biology first. Mitochondria wear out, and the cell has a system for clearing the damaged ones so the network can renew: mitophagy. It weakens with age, and that weakening is consequential. In worms and old rodents, urolithin A switched the system back on and muscle function improved. Most people cannot make the molecule reliably from pomegranate; in a study of a hundred Americans, only about forty percent converted meaningfully and a third could not convert at all. Five hundred milligrams of Mitopure gave everyone six times the exposure of a glass of juice. That part is settled, and it is genuinely clever.
“Then the human question, which is the only one the Standard cares about. Take Mitopure for four weeks and a signature appears in the blood and the muscle: acylcarnitines fall, mitochondrial gene sets shift. That is a signature of the mechanism, not a photograph of it. Nobody has watched human mitochondria being recycled, and in the one trial that measured how much ATP the muscle could make, it did not change.
“Function is where I read most carefully. In sixty six adults aged sixty five to ninety, four months at a thousand milligrams did not significantly improve the six-minute walk or ATP production, the two endpoints the trial was built to test. Endurance in the hand and leg improved at two months and had faded by four. In eighty eight middle-aged adults at the dose sold, peak power, the primary endpoint, did not change; hamstring strength rose twelve percent against placebo, quadriceps did not, and the higher dose did less than the lower one. The company's label says up to twelve percent stronger. That is true of one muscle, at one dose, in sedentary overweight middle-aged adults, as a secondary outcome, and an advertising review made them say so.
“Then the syntheses, which are why this file will not be an advertisement. An independent systematic review in 2024 found inflammation eased, some genes moved, strength and endurance up, and no effect on ATP, mitochondrial biogenesis, body measures, heart outcomes or physical function. In July 2026 a meta-analysis, with a co-discoverer of the mechanism among its authors, pooled the only outcome it could, the six-minute walk: seventeen metres in favour, a confidence interval that crosses zero, low certainty. Their words: exploratory and hypothesis-generating, not reproducible evidence of efficacy. The signal is increasingly difficult to dismiss. It is not yet consistent enough to call definitive.
“The safety record is a genuine credit. No product-related adverse event in any trial at the dose sold or at twice it, a ninety-day toxicology study with nothing found at the highest dose, and a GRAS letter from the FDA with no questions. That letter also says, in its own words, that it is not an affirmation, and it covers oatmeal and yogurt, not softgels. The website's badge reads FDA GRAS Favorable Review and the FAQ says the FDA declared it safe. The letter says less than that, and so does this file.
“The softgel itself is well made and easy to live with: two a day, tasteless, no refrigeration, a subscription that matches the trial length. It is NSF Certified for Sport, which means someone outside the company checks each lot for what is on the label and what should not be in it. It also contains annatto, a colourant a clean formula did not need, and Timeline's own label image does not list the excipients at all; I read them from a retailer.
“And then the pages, which a company this careful about its trials has not been careful about. Purity is ninety seven percent minimum in the specification the FDA read, ninety eight on the ingredient's site and ninety nine in the FAQ. Patents are forty, fifty, eighty or a hundred depending on where you look. Human trials are twenty five on the homepage, seven in the FAQ, five on Amazon. The container is a sachet on one page and a bottle on another. A company article describes an EFSA authorisation that has not happened. None of this is about the softgel. All of it is about trust.
“So, the anchors. Institutional score 3.52. The floor is Stewardship at 2.71: nothing published about the container, its material, its recyclability, a carbon figure or a life-cycle position. Efficacy sits at 3.71, which is high for a supplement and honest about a signal. The computation is Finalist. The founder ruled One Bio Star, for the molecule, the dose and the programme, and the file prints both numbers.
“What would move it. One page about the object would lift the floor. One set of figures used on every page would lift trust. A six-month trial at the dose sold is already recruiting; if the hamstring result repeats and the walk result stops crossing zero, Efficacy moves, and with it the file.
“Mitopure has already crossed a threshold that few longevity supplements reach: the mechanism has left the diagram and entered randomised human investigation. What remains unresolved is whether those molecular and functional signals become sufficiently consistent, durable and consequential to alter the trajectory of human ageing. The science has travelled unusually far. It has not reached its final destination.”

13-Circle Findings
Circle by Circle.
I · Core Integrity
Pass
Pass, flagged in four places. The molecule, the dose and the purity are as claimed, the trials are published with their missed primaries, and no enforcement action exists. Flagged: three purity figures, four patent counts and three trial counts across the company's own pages; a GRAS letter described as a declaration of safety when it calls itself not an affirmation; an EFSA authorisation described in a company article that has not happened; and a set of muscle claims the National Advertising Division narrowed in 2025.
II · Efficacy
Pass
3.71. Exceeds the standard on evidence base, mechanism and integration, and is the exemplar on formulation rigor: the five hundred milligram serving sold was itself randomised for sixteen weeks. Held at Meets on consistency, clinical significance and independent validation: both major trials missed their primary endpoints, the strength signal is one muscle in one trial, the pooled walk effect is inconclusive at low certainty, and the independent syntheses call the functional findings exploratory.
III · Physiological Alignment
Pass
3.67. Biological in method: the body's own metabolite, supplied to those whose microbiome does not make it, with no stimulant and no pharmacological lever. Resilience over years is asserted; no trial runs past four months.
IV · Regulation & Recovery
Pass
3.60. Low physiological burden, no product-related adverse events at the dose sold or at twice it, a toxicology programme with nothing found at the highest dose, and population language that keeps pregnancy and children behind medical advice.
V · Input Quality
Partial
3.43. A characterised synthesised molecule released against a specification the FDA has read, in a plant-based softgel. Held below 3.50 because the company states purity three ways, publishes no certificate of analysis, adds a colourant a clean formula did not need, and omits the excipient list from its own label image.
VI · Environmental Quality
Pass
3.50. A plant-based shell, a triglyceride carrier and lecithin; a tasteless softgel; a container whose material is not stated and whose name changes between pages. Nothing questionable and nothing distinguished.
VII · Operational Precision
Pass
3.50. Process rigor is strong: chemical synthesis with a filed specification, cGMP and FSSC 22000, and NSF Certified for Sport on the exact softgel with published lot numbers. One hard mark holds the Circle to 3.50: reliability across touchpoints, where the company's pages disagree with each other on purity, patents, trials, the container and the meaning of its GRAS letter.
VIII · Care & Competence
Pass
3.57. Founded and led by scientists, with the discovering laboratory on its advisory board and a chief medical officer who publishes the trials under his own name. Guidance to the customer is clear and, since 2025, precisely qualified. Held below Exceeds by conventional service standards and by the fact that the same people design, fund and analyse the trials.
IX · Protocol Design
Pass
4.00. The one Circle at 4.00. Two softgels a day, nothing to learn, a subscription cadence that matches the trial, a sample, plain pricing. The deduction is trust architecture: a badge and a set of counts the pages do not agree on.
X · Aesthetic Restraint
Pass
3.57. Discipline in the formula and the visual system, one colourant it did not need, and a voice that runs well ahead of the evidence: clinically proven to rejuvenate mitochondria, root causes of aging, twenty five human trials.
XI · Stewardship
Partial
2.71, the floor of the file. A synthesised molecule that takes no orchard and a refill format for the powder are real. Nothing is published about the softgel's container, its material, its recyclability, a carbon figure or a life-cycle position.
XII · Cultural Responsibility
Partial
3.29. Attribution of the science is a strength: every trial names its funder and its employee authors. Manufacturing, sourcing and labour are described by country and standard, not by name.
XIII · Enduring Value
Pass
3.71. On the market since 2020 with the same molecule at the same dose, published with its nulls, with a six-month trial at the dose sold now recruiting. Repeatable at the level of exposure and signature; not yet at the level of function; durability untested beyond four months.
The Record
What was examined.
The molecule
Urolithin A, a metabolite of ellagitannins produced by certain gut bacteria, not present in food itself.
The origin
Ellagitannins and ellagic acid in pomegranate, walnuts and some berries; conversion by intestinal microbes into urolithins.
The variability
In one hundred American adults, twelve percent had detectable urolithin A before pomegranate juice, about forty percent converted meaningfully after it, and a third could not convert at all. Conversion capacity varies with age, microbiome and geography.
The intervention
Supply the metabolite directly at a standardised dose rather than rely on the microbiome to make it.
Mitopure
Amazentis's proprietary, chemically synthesised urolithin A. Specification filed with the FDA: minimum ninety seven percent purity, with limits on cadmium, lead, arsenic, mercury, solvents and microorganisms, stable three years at room temperature. Stated as more than ninety eight percent on the ingredient's sites and more than ninety nine in the FAQ.
The dose
Five hundred milligrams a day in two softgels, the dose randomised for sixteen weeks in the middle-aged trial. One thousand milligrams in the older-adult trial and the immune and athlete trials.
The exposure
Crossover study of one hundred adults: five hundred milligrams of Mitopure produced six times the urolithin A exposure of eight ounces of pomegranate juice by area under the curve over twenty four hours, and 2.4 times at the twenty-four-hour point. Funded by Amazentis.
The mechanism
Mitophagy, the selective removal of damaged mitochondria. Induced by urolithin A in worms and rodents, with lifespan and muscle-function effects there. In humans, a molecular signature: plasma acylcarnitines fall and mitochondrial gene sets in muscle shift after four weeks; maximal ATP production unchanged where measured.
The first human signal
Nature Metabolism, 2019. Sixty adults aged sixty one to eighty five; single doses to two grams and four weeks at two hundred and fifty, five hundred and one thousand milligrams; safety the primary outcome, met; acylcarnitines and muscle mitochondrial gene expression modulated at five hundred and one thousand milligrams. Amazentis-funded, employee authors.
The older-adult trial
JAMA Network Open, 2022. Sixty six adults aged sixty five to ninety, one thousand milligrams for four months. Primary endpoints, six-minute walk distance and maximal ATP production in hand and leg muscle, not significantly improved. Secondary: hand and leg endurance improved at two months, not at four; acylcarnitines and ceramides reduced; CRP eased. Adverse events no different. Amazentis funded and had a role in design, analysis and the decision to publish.
The middle-aged trial
Cell Reports Medicine, 2022. Eighty eight adults aged forty to sixty four, five hundred or one thousand milligrams for sixteen weeks. Primary endpoint, peak power output, not significantly changed. Hamstring peak torque plus twelve percent against placebo at five hundred milligrams and plus 9.8 at one thousand; knee flexion plus ten percent; quadriceps and grip unchanged. Acylcarnitines and muscle gene sets moved at five hundred and not at one thousand. Amazentis funded.
The systematic review
Ageing Research Reviews, 2024. Five studies, two hundred and fifty people, ten to one thousand milligrams, twenty eight days to four months: anti-inflammatory effect, some mitochondrial and autophagy genes up, fatty-acid oxidation up, strength and endurance up; no effect on maximal ATP production, mitochondrial biogenesis or dynamics, body measures, cardiovascular outcomes or physical function. Independent.
The 2026 meta-analysis
Frontiers in Nutrition, July 2026. Five randomised trials, two hundred and thirty six people. Six-minute walk pooled: plus 17.03 metres, 95 percent CI minus 5.33 to 39.40, P 0.135, low certainty. Strength, endurance and biomarker findings exploratory and hypothesis-generating, not reproducible evidence of efficacy. No commercial funding; senior author a co-discoverer of the mechanism.
The strength signal
Twelve percent in hamstring peak torque against placebo, at five hundred milligrams, in sedentary overweight adults aged forty to sixty four, as a secondary outcome of one trial. Qualified to exactly that by the National Advertising Division in May 2025.
The energy claim
Increases cellular energy rests on acylcarnitine and gene-expression signatures. Maximal ATP production did not change in the one trial that measured it.
The safety record
No product-related adverse events in any trial; ninety-day rodent study with the no-effect level at the highest dose tested; not genotoxic. Not studied in pregnancy, lactation or children, and the company says so.
The regulatory record
GRAS notice 791, FDA letter of 20 December 2018: no questions regarding the company's conclusion for specified food uses; not an affirmation. EU novel-food application of 2018 still in risk assessment. No FDA or FTC action. National Advertising Division decision, May 2025.
The purity
Minimum ninety seven percent by filed specification; more than ninety eight and more than ninety nine percent by company pages; no batch certificate published.
The manufacturing
Ingredient and finished product made in the United States under cGMP; FSSC 22000, ISO 22716 and ISO 9001 stated for the ingredient; facilities unnamed; country of synthesis unstated.
The certifications
NSF Certified for Sport on the softgel, with published lot numbers, testing label content and contaminants. Clean Label Project listing; scope not confirmed. Vegan and non-GMO self-declared.
The conflicts
Discovery, exposure, phase one, both randomised trials and the immune trial funded by Amazentis with employees and board members as authors, all disclosed. Systematic review, meta-analysis and two small trials independent, one of the trials single-blind with travel reimbursed by the brand.
The softgel
Two plant-based softgels a day, tasteless, with or without food, on the larger side. Excipients a vegan shell, MCT oil, lecithin and annatto, per a retailer's transcription; not on Timeline's own label image.
The price
Ninety nine dollars for thirty days at list, three dollars thirty a day; sixty nine to eighty nine on subscription. Behind it: a proprietary synthesised molecule, a filed specification, NSF certification and a fifteen-year research programme.
The limit
Institutional score 3.52, floor 2.71 at Stewardship, one Circle at 4.00: computed Finalist. A Star earned outright needs the container and footprint described and the figures reconciled; a second Star needs the functional signal to repeat at the dose sold; a third needs consequence.
